Here are currently the only confirmed characters for Twilight Princess.

Colin

Another Ordon villager, Colin is a young boy who, like Ilia, is very in tune with nature. He is apparently distant in human relations. He, like Ilia, is captured and must be rescued by Link at some point in the game.

Fado

Fado was the deceased Wind Sage in The Legend of Zelda: The Wind Waker. While his current role in the game (if any) is unknown, rumors have been circulating around the internet regarding his presence in the game, as he is apparently mentioned by the Mayor.

Ganon

Although Ganon has been confirmed to be in the game, currently his role in it is unknown. According to Nintendo, he will not be taking up his traditional role in the game, which really leaves the window open for anything.

Hena

Hena is a young woman who owns the Fishing Hole. Hena inherited her job from an ancestor of hers, who ran the fishing pond on the shore of lake Hylia many ages ago.

Link

Link is the hero of Twilight Princess. He begins the game as a wrangler in Ordon Village. He is summoned by the mayor to attend the Hyrule Summit, but is suddenly transformed into a wolf once he is exposed to the Twilight Realm.

llia

Ilia is one of Link's closest friends in Ordon Village. She is the daughter of the Mayor. She is very connected with nature and has special healing abilities. She is captured, along with Colin, at some point in the game, causing Link to come to her rescue.

Mayor

The Mayor of Ordon Village, and Ilia's father. It is the mayor who selects Link to attend the Hyrule Summit as the representative of Ordon Village.

Midna

Midna seems to be the most mysterious known character in Twilight Princess. She is seen riding Wolf Link through his first voyage through the Twilight Realm, but aside from that, nobody is sure of where her true allegiance or goals lie. Rumor has given rise to the belief that Midna is evil, but is working together with Link to stop a much greater threat. Only time will tell...

Ordona

Ordona, the Light Spirit of the Ordona Province, plays a vital role in Link's quest to restore light to Hyrule. In order to restore light to an area, you'll need to collect Tears of Light. Ordona will then present Link with the Vessel of Light, in which the collected tears will be placed. Only when you do that will light be restored to the area.

Wolf Link

When Link enters the Twilight Realm, he is transformed into Wolf Link. Upon transforming, he is captured by some unknown force, and is forced to form an alliance with Midna in order to escape. It is currently unknown how long Link remains in his wolf form, and how long Midna remains an ally.

Zant

Zant, a mysterious man in black robes is seen being escorted by two twilight enemies down a dark hall. It seems that he is the Twilight King, or at least an antagonist that holds a lot of importance to the Twilight Realm.

Zelda

Zelda has been confirmed to be in Twilight Princess, but, like Ganon, her role in this game is different from her traditional one. When the official artwork for Zelda had her weilding a sword, rumors began circulating that Zelda is a playable character. Whether or not that is possible is currently unconfirmed. What we do know is that Zelda spends some portion of the game in a dark tower (possibly imprisoned), where she is seen in various screenshots wearing a long hooded cloak. According to Nintendo, this cloak is a magical device which Zelda uses to protect herself from monsters.
