Skulltulas are large spiders that drop down from the ceiling. They are armored, so if they are hanging on their web, wait until they turn around before you slash away. If they are on the ground, like in Twilight Princess, you must catch them off guard by attacking suddenly and unpredictably, or by using a Shield Attack. Otherwise, they will block all sword attacks with their front legs. There is also a smaller species of Skulltula that hang on walls, and are aptly names Skullwalltulas, although they are also called Skulltulas quite often.
